? Happy #FluorescenceFriday! ? Check out this stunning 3D video of human normal adjacent colorectal cancer FFPE tissue, straight from our SITC poster! For the technical experts, here’s the breakdown: ?? Tissue Prep: FFPE tissue was deparaffinized and processed, then stained with nuclear dye YO-PRO-1 (blue) and an anti-CD8 antibody (yellow). ?? Optical Clearing: Modified iDISCO+ protocol ?? Imaging: 2 μm/pixel resolution using the hybrid open-top light-sheet microscope from Alpenglow Biosciences (the 3Di). ?? Analysis: CD8+ lymphocytes were quantified in the entire tissue section (approx. 6 mm x 4.3 mm x 4.2 mm) using advanced 3D spatial analysis tools. What makes 3D imaging a game-changer here? ?? It enables us to clearly visualize crypt morphology in colorectal tissue, offering an unmatched view of the structural details vital to understanding tissue organization and pathology. ?? Lymphocyte density varies dramatically depending on section depth, which you can see starting at sec 14 as we virtually "section" through the tissue. ?? This variability highlights the power of 3D imaging to uncover insights that traditional 2D sections can’t reveal. Want more data and insights? ?? Discussing breakthroughs from the Human Tumor Atlas Network (HTAN) and the recent publication -Spatial Awareness- https://lnkd.in/e-p-Rnws ?? Through 3D mapping at the single-cell level, scientists are reshaping approaches to cancer diagnosis, treatment, and prevention. Nature’s “Spatial awareness” collection highlights how detailed 3D cellular mapping unveils tumor structures across various cancers, enabling researchers to pinpoint each cell's location and molecular state within tumors. This approach reveals critical insights into how cancers evolve and interact with surrounding tissue. HTAN’s creation of cellular maps for breast, pancreatic, and colon cancers has underscored a crucial finding: tumors are not uniform. Different regions within a single tumor can respond uniquely to treatments, demonstrating the need for highly targeted therapies tailored to each region's specific molecular makeup. Additionally, 3D mapping reveals the earliest molecular shifts that can indicate cancer risk. For instance, researchers observed DNA and gene activity changes associated with immune responses and hormone regulation in pre-cancerous lesions like colon polyps. Detecting these early markers presents an opportunity for prevention, as doctors could intervene before cellular changes turn irreversible. For immunotherapy, 3D mapping reveals valuable insights impossible with classical 2D technologies. In breast cancer, which often resists immunotherapies, 3D mapping shows that immune cells are distributed unevenly. Some tumor areas contain "exhausted" immune cells that are ineffective in combatting cancer. This information helps scientists understand why tumors can evade immune-based treatments and points toward strategies to boost immunotherapy’s success against resistant cancers. In colorectal cancer research, scientists have used CRISPR gene-editing to establish a “molecular clock” that tracks cellular changes over time. This method has shown that many colon polyps are more complex than previously thought, often involving multiple cell types rather than stemming from a single mutated cell. As a result, assessing cancer risk may require new biomarkers that capture genetic changes over time, revealing tumors’ behavior beyond just size. ?? These 3D mapping advances are opening a new chapter in cancer research, transforming our understanding of how tumors function, evolve, and can be targeted. By viewing cancer in previously unseen detail, scientists can now track cellular contributions to tumor growth and identify intervention points, offering hope for more precise, early, and individualized treatment approaches.
