As a public speaker it’s a challenge that needs to be addressed. First of all set the boundaries and ground rules that a separate time has been earmarked for questions and answers. You can note the question and answer only in the end of the session/ discussion. Ask your audience to keep their questions ready for discussion after you have finished.?

Refocus the audience to get back on track. Always be mindful of time. Acknowledge that the discussion can be discussed in full at a later time. That is something I have observed listed companies' Board Chairpersons doing it well. The key is to address the emotions or the feelings of the audience member who initiated the interruption. In a large gathering, it takes courage for the person to speak up, hence that person must have a strong (emotional) reason to raise the issue, i.e. the issue is something the person feels strongly about. The "Think-Act-Feel" framework is a useful reminder that questions and issues are not just on facts and logic but can be based on emotional impetus from the persons speaking up.

Use framing in your planning. Solution? At the beginning of the presentation clearly outline that you require participants to actively listen, suggest that each time a question(s) come up, write it down, then once the speaker has finished review the list of questions and then, there is an opportunity to ask them. The speaker maintain flow, assisting with the context and story. I feel this approach frames and delivers respect for all involved.

Establishing ground rules is useful and the clearest ground rules is whether the talk or presentation is meant to be interactive where interruptions are welcomed or it is more formalized that questions should only come during the question and answer segment. If the session is a moderated panel or there is a moderator, the moderator could play this role to set the ground rules to balance between the flow of the talk and promoting audience engagement with interactive questions or comments.

Sometimes it can help to draw a physical "parking lot" for ideas and questions to come back to. This helps to establish ground rules but also gives the audience comfort that they will be given time. So, draw/write them on a whiteboard, or ask for questions to be posted in the chat etc

I learnt from an experienced trainer that even if you lose your train of thought (which WILL happen!), you can use the audience as a resource. I have sometimes asked the audience where was I in terms of my presentation before I digressed to address an interruption due to an urgent question from an audience member. The audience helpfully reminded what I the point I was at before I was interrupted.

This politely signals that you need to maintain focus on your content. To echo what Eh-Chaun Yoong is suggesting below, sometimes the conversation rambles for few minutes until it reaches a point that is a kind of far from the main topic. I smile and ask the audience: "Where was the starting point? how did we get here?". I always have one of the audiences who is able to hold the beginning of the thread of the conversation.

Humor is a double-edged tool, it can help but it can hurt (your objectives) if it is not appropriate to the audience and topic. After participating in many humorous speech contests during my ongoing Toastmasters journey, I have often struggled to deliberately inject humor into my speeches. I write something that I think is funny, yet, when the speech is delivered, the audience barely releases a giggle. The key to successful humor is to be able to read the pulse of the room and to understand what are the demographics of your audience. I have found this easy in theory but difficult in practice. Everyone has a unique taste in humor, the trick is to find the common denominator that releases the tension (if any).

Lines like: "What do you know, that's actually on the next slide!" "Have you seen this presentation already, you seem to know all the bits that are coming up" Gentle enough to admonish the interruptor without offending, but also helpful in getting the rest of the audience on your side.
