Defining properly a problem is critical. As Marvin Bower said "the most frequent cause of failures in business is not people who answered the right questions incorrectly, but people who answered the wrong questions correctly". Therefore the art of defining a problem is the art of raising the right questions. Then in the biotechnology field as in any other field, the problem needs to be broken down in smaller issues than will subsequently be prioritized.

Before going to all those steps, sometimes a problem needs creativity and therefore a proper brainstorming approach. Brainstorming can be conducted with different frameworks but it shouldn't be solution oriented. The goal is to find all the good ideas related to the problem even before defining it properly. Consider adding members that are not closed to the project to have an unbiased view on the problem.
